Is a reboot required in NT 4.0 for a new or modified entry
to the HOSTS file?

Thanks.

This is pretty easy to determine on your own.  Open a command prompt
(cmd.exe) and enter this:

echo TESTHOSTS 127.0.0.1>>%windir%\system32\drivers\etc\hosts
ping TESTHOSTS

Does it resolve?
